Product Use

Used to determine the activity of glutamate dehydrogenase (GLDH) in human serum and plasma.

Glutamate dehydrogenase is a mitochondrial enzyme present in all tissues. in vivo GLDH has the highest content in the liver, mainly in hepatic lobular cells, and the enzyme activity is extremely low in normal serum. gLDH is a liver-specific mitochondrial enzyme that enters the blood when hepatocytes are necrotic, and the level of serum GLDH can reflect the degree of hepatocyte damage.

Detection Principle

GLDH

α-ketoglutarate + NH3 +NADH ───→ Glutamic acid +NAD+

The rate of change of NADH absorbance per minute was measured at 340 nm and its content was calculated.

Storage Expiration Date

For opening the kit can be stable for one year when stored at 2~8℃ away from light; reagents can be stable for one month when stored at 2~8℃ away from light after opening the bottle. Reagents should not be frozen.

Sample Request

For serum or heparin/EDTA anticoagulated plasma specimens, the serum should be separated promptly after blood collection to avoid hemolysis. Specimens should be separated by centrifugation immediately after blood collection, and samples can be stored at room temperature (15-25°C) for 7 days, 2-8°C for 7 days, and -20°C for 4 weeks.

  1. QC control procedures.

It is recommended that each laboratory establish its own quality control system and select appropriate quality control products for quality control. The measured values of QC products should be within the specified range. If outside the specified range, it is necessary to take appropriate measures or contact the manufacturer.

  1. Calculation.

GLDH activity (U/L) in the sample = (ΔAsample /min-ΔAblank /min) × Cstandard / (ΔAstandard /min-ΔAblank /min).

Reference Range

Serum and plasma: Adult male 0-7 U/L Adult female: 0-5 U/L

It is recommended that each laboratory establish its own reference range of normal values.

Anti-interference

Ascorbic acid ≤ 40 mg/dl, bilirubin ≤ 40 mg/dl, hemoglobin ≤ 500 mg/dl, triglycerides ≤ 2000 mg/dl did not interfere with the determination.

Performance Indicators

Absorbance of reagent blank: wavelength 340 nm, optical diameter 1.0 cm, temperature 37°C± 1°C, A0  ≥ 0.8. A0 < 0.8 when disabled or replaced with new reagent.

Reagent blank absorbance change rate: wavelength 340 nm, optical diameter 1.0 cm, temperature 37 ℃± 1 ℃, the measured reagent absorbance change value ︱ △A/min ︱ ≤ 0.0005.

Analytical sensitivity: the kit tests 10 U/L of the test substance, the absorbance change rate ΔA/min ≥0.001.

Linearity range: 2~120 U/L (Judgment basis: r ≥ 0.990). If out of linear range, please measure after dilution with saline and multiply the result by the dilution factor.

Precision: intra-batch coefficient of variation ≤ 6.0 %; inter-batch coefficient of variation ≤ 8.0 %.

Accuracy: Absolute value of relative deviation ≤ 10 %.

Correlation: Comparison with similar products: n = 120, r = 0.9941, y = 0.9738x + 0.3936 (y: our reagent; x: other reagents)

6, the theoretical K value is for reference only, and the actual K value may not be the same, so it is recommended to use the calibration serum to set the standard, high fat or jaundice samples should be diluted and then tested.
