Intended Use  

In vitro determination of Mitochondrial Aspartate aminotransferase(mAST) in human serum

(mAST) activity.

Aspartate aminotransferase (AST) is widely present in many organs, including heart, liver, skeletal muscle, and kidney in order of abundance. two isozymes of AST, controlled by different genes, are present in the cytoplasm (cAST) and mitochondria (mAST). mAST is an isoenzyme of aspartate aminotransferase located in the mitochondria, and the release of the mitochondrial enzyme is higher in severe tissue injury. This can lead to an increase in serum mAST. An increase in serum mAST reflects the severity of subcellular structural damage and is a sensitive indicator for the diagnosis of hepatocellular damage and myocardial infarction. In liver disease, mAST can be used to evaluate hepatocyte necrosis and determine healing, and to diagnose patients with active alcoholic liver disease; in heart disease, it can determine the effectiveness of treatment and healing of myocardial infarction.

Test Principle

This method is based on the determination of mAST activity in the presence of a protease that completely inhibits the activity of cAST without affecting the activity of mAST, and subsequently using a method for measuring total AST activity.
